lebrikizumab

EbglyssIL-13 inhibitor

Ebglyss (lebrikizumab) is a monoclonal antibody from Eli Lilly that selectively neutralises interleukin-13, a key driver of type 2 inflammation in atopic dermatitis. It is used in moderate-to-severe disease in adults and adolescents when topical treatment is not enough.

This page is based on FDA prescribing information and the EMA medicine record for lebrikizumab, cross-checked with DailyMed and the National Cancer Institute dictionary; the mechanism is explained in plain language, and analogies are used only where they stay accurate.

How it works

Explains what the medicine does in the body, not dosing, side effects, or safety.

An analogy

Atopic dermatitis is like a faulty alarm circuit that keeps the skin itchy and inflamed, and IL-13 is one of the main wires carrying the alarm. Lebrikizumab clips that specific wire. Other wires, including IL-4's, stay intact, but the alarm's volume drops enough for the skin barrier to repair and the itch to settle.

In simple terms

In atopic dermatitis the immune system overproduces inflammatory signals, and IL-13 is one of the most important. It promotes itching, weakens the skin barrier and keeps inflammation going. Lebrikizumab binds IL-13 and prevents it from activating its receptor. With that signal blocked, itch and redness decrease and the skin has a chance to heal.

In depth

Lebrikizumab is a humanised IgG4 monoclonal antibody that binds IL-13 with high affinity, selectively preventing formation of the IL-13/IL-4R-alpha/IL-13R-alpha1 signalling complex while sparing IL-13R-alpha2 and IL-4 signalling. This suppresses type 2 inflammatory pathways, including IgE production and epithelial barrier dysfunction. Trials demonstrated sustained skin clearance and itch improvement with dosing that can be extended to every four weeks after induction.

Target: Interleukin-13 (IL-13)

A type 2 cytokine central to the itch, barrier disruption and inflammation of atopic dermatitis; blocking it calms the allergic inflammatory loop without touching IL-4 signalling.
